Identification of Albizia lebbeck seed coat chitin-binding vicilins (7S globulins) with high toxicity to the larvae of the bruchid Callosobruchus maculatus BJMBR
Souza,A.J.; Ferreira,A.T.S.; Perales,J.; Beghini,D.G.; Fernandes,K.V.S.; Xavier-Filho,J.; Venancio,T.M.; Oliveira,A.E.A..
Seed coat is a specialized maternal tissue that interfaces the embryo and the external environment during embryogenesis, dormancy and germination. In addition, it is the first defensive barrier against penetration by pathogens and herbivores. Here we show that Albizia lebbeck seed coat dramatically compromises the oviposition, eclosion and development of the bruchid Callosobruchus maculatus. Dietary supplementation of bruchid larvae with A. lebbeck seed coat flour causes severe weight loss and reduces survival. By means of protein purification, mass spectrometry and bioinformatic analyses, we show that chitin-binding vicilins are the main source of A. lebbeck tegumental toxicity to C. maculatus. At concentrations as low as 0.1%, A. lebbeck vicilins reduce...

Lima bean (Phaseolus lunatus) seed coat phaseolin is detrimental to the cowpea weevil (Callosobruchus maculatus) BJMBR
Moraes,R.A.; Sales,M.P.; Pinto,M.S.P.; Silva,L.B.; Oliveira,A.E.A.; Machado,O.L.T.; Fernandes,K.V.S.; Xavier-Filho,J..
The presence of phaseolin (a vicilin-like 7S storage globulin) peptides in the seed coat of the legume Phaseolus lunatus L. (lima bean) was demonstrated by N-terminal amino acid sequencing. Utilizing an artificial seed system assay we showed that phaseolin, isolated from both cotyledon and testa tissues of P. lunatus, is detrimental to the nonhost bruchid Callosobruchus maculatus (F) (cowpea weevil) with ED50 of 1.7 and 3.5%, respectively. The level of phaseolin in the seed coat (16.7%) was found to be sufficient to deter larval development of this bruchid. The expression of a C. maculatus-detrimental protein in the testa of nonhost seeds suggests that the protein may have played a significant role in the evolutionary adaptation of bruchids to legume seeds.

Seed morphology and classification of Impatiens (Balsaminaceae) Naturalis
Utami, N.; Shimizu, T..
Seed coat morphology was observed through SEM for 65 species and 10 sections of Impatiens. Several types of seed coat are recognized in this study. On the basis of comparison with other characters such as chromosome numbers, floral characters and seedling morphology, systematic problems concerning Impatiens are briefly discussed.

Seed morphology of some tribes of Brassicaceae (implications for taxonomy and species identification for the flora of Egypt) Naturalis
Khalik, K. Abdel; Maesen, L.J.G. van der.
Seed morphology of 45 taxa belonging to 23 genera of Brassicaceae were examined using light and scanning electron microscopy. The taxa included representatives of the tribes Arabideae, Euclidieae, Hesperideae, Lunarieae, Matthioleae, and Sisymbrieae, which all occur in Egypt. Macro- and micromorphological characters, including seed shape, colour, size, the position of the radicle relative to the cotyledons, epidermal cell shape, anticlinal boundaries, outer periclinal cell wall and relief of outer cell walls, are presented. Three types of basic anticlinal cell wall boundaries are recognised and six different shapes of the outer periclinal cell wall are described. A key for the identification of the investigated taxa based on seed characters is provided.

Testa structure in Erythrina speciosa (Leguminosae): the role of the mucilaginous stratum in the acquisition of physical dormancy Acta Botanica
Magalhães,Camila Ribeiro; Oliveira,Denise Maria Trombert.
ABSTRACT Seeds of Erythrina speciosa (Papilionoideae) exhibit physical dormancy, but evidence has suggested that the proportion of impermeable seeds increases with storage. There are no records of structural variation in the testa of E. speciosa that justifies the difference in the percentage of seeds with physical dormancy. Therefore, the present study investigated the difference in permeability between mature seeds of E. speciosa that were freshly collected and those stored for 12 months. Seeds were examined for changes in testa structure that could result in differences in the percentage of seeds in physical dormancy. Thus, imbibition curves and water gaps were determined, and surface micromorphology, anatomy, and histochemistry of the testa were...

The role of the seed coat in the light sensivity in Raphanus sativus L. cv. redondo gigante seeds Acta Botanica
Gonçalves,Maura Lúcia Costa; Takaki,Massanori.
The role of the seed coat in the light sensitivity of seeds of Raphanus sativus L. cv. redondo gigante was analysed by germination tests of intact and naked seeds. Far-red light caused high inhibition of seed germination, while under white and red lights low inhibition was found. Naked seeds presented no light sensitivity with high percentage germination under light and darkness. However, incubation of naked seeds in -0.6MPa polyethylene glycol solution resulted in light inhibition as observed in intact seeds. The analysis of the seed coat transmitted light indicated that the filtered light presented the same photoequilibrium of phytochrome when compared to the white light, with a decrease of only 33% in the light irradiance which reaches the embryo.
